Determination of Secoisolariciresinol Diglucoside SDG in Callus of Linum usitatissimum L. exposed to heat and cold shock by HPLC

Abstract
Callus produced from cotyledon cultured on MS medium supplemented with 0.25 mg.L-1 NAA and 0.4 mg.L-1 BA after 4 weeks divided into 1g and cultured in the same medium was exposed to heat shock for two times, 10 or 20 minutes at 40 45 and 50° C and cold shock at 0° C for 5, 10, and 15 minutes then determine the quantity of SDG by HPLC. The data refers that exposing callus to 10 minutes at 0° C gave the highest fresh weight and protein 14.49 g and 27.51% respectively, and the treatment achieved the highest area under the curve and highest SDG content 1346.641 and 37.88 mg.g-1 respectively, otherwise the Highest SDG content 33.28 mg.g-1 has been obtained from exposing callus to 50° C for 20 minutes.
